چکیده

The binding of GnRH to its receptor results in the synthesis and secretion of the gonadotropins, as well as stimulation of the gene encoding its own receptor. Thus, the interaction between GnRH and GnRHR represents a central point for regulation of reproductive function. Glucocorticoids can alter reproduction by reducing GnRH responsiveness of gonadotropes within the anterior pituitary gland, potentially via transcriptional regulation of the GnRHR gene. Investigation of this possible mechanism, however, revealed that transcription of the murine GnRHR gene is stimulated by glucocorticoids. To determine the effect of glucocorticoids on porcine GnRHR gene expression, gonadotrope-derived αT3-1 cells were transiently transfected with a vector containing 5118 bp of 5’ flanking sequence for the porcine GnRHR gene fused to luciferase for 12 h and treated with increasing concentrations of the glucocorticoid agonist, dexamethasone (0, 1, 10, 100 and 1,000 nM) for an additional 12 h prior to harvest. Maximal induction of luciferase activity was detected at 100 nM of dexamethasone (2-fold over vehicle; P < 0.05), and this response was blocked by the glucocorticoid antagonist, mifepristone (100 pM). Deletion from 274 to 323 bp upstream of the translational start site eliminated glucocorticoid responsiveness, suggesting the presence of a GRE(s) within this region. Structures, Functions and Expressions of GnRH and GnRH Receptor in Peripheral Reproductive Organs and Their Regulation by Estradiol-17β

Studies have shown that estradiol-17β (E2) regulates gonadotropin-releasing hormone (GnRH) and GnRH receptor expression in hypothalamus and pituitary. Several studies have shown that GnRH and its receptor are also expressed in peripheral reproductive organs and little is known about their regulations. In this study, GnRH and GnRH receptor structures, functions, their peripheral expressions and ...

Molecular biology of gonadotropin-releasing hormone (GnRH)-I, GnRH-II, and their receptors in humans.

In human beings, two forms of GnRH, termed GnRH-I and GnRH-II, encoded by separate genes have been identified. Although these hormones share comparable cDNA and genomic structures, their tissue distribution and regulation of gene expression are significantly dissimilar. The actions of GnRH are mediated by the GnRH receptor, which belongs to a member of the rhodopsin-like G protein-coupled recep...

Activity of the porcine gonadotropin-releasing hormone receptor gene promoter is partially conferred by a distal gonadotrope specific element (GSE) within an upstream enhancing region, two proximal GSEs and a retinoid X receptor binding site

BACKGROUND Regulation of gonadotropin-releasing hormone (GnRH) receptor (GnRHR) numbers on gonadotropes within the anterior pituitary gland represents a critical point for control of reproductive function. Binding of GnRH to its receptor regulates follicle stimulating hormone (FSH) and luteinizing hormone (LH) release and levels of this G-protein coupled receptor on the surface of gonadotropes ...

Differential use of signal transduction pathways in the gonadotropin-releasing hormone-mediated regulation of gonadotropin subunit gene expression.

The regulation of LH and FSH subunit gene expression is under the control of GnRH. Physiological changes in the frequency of pulsatile GnRH release from the hypothalamus result in differential stimulation of alpha-, LHbeta-, and FSHbeta-gene expression. Previous studies indicate that the GnRH receptor couples to G proteins of the G(q/11) family, with phosphoinositide turnover and its resultant ...

Regulation of miR159 and miR396 mediated by Piriformospora indica confer drought tolerance in rice

Drought stress is one of the most determinative factors of agriculture and plays a major role in limiting crop productivity. This limitation is going to rising through climate changes. However, plants have their own defense systems to moderate the adverse effects of climatic conditions. MicroRNA-mediated post-transcriptional gene regulation is one of these defense mechanisms. The root endophyti...
